In order to find function of TTHA0344 protein from T. thermophilus HB8, we observed the expression profile of the total mRNA of the TTHA0344-deficient strain grown in a rich medium at 70°C for 680 min. Overall design: Three TTHA0344-deficient T. thermophilus HB8 strains were each pre-cultured at 70°C for 16 h in 3 ml of TT medium containing 0.8% polypeptone, 0.4% yeast extract, 0.2% NaCl, 0.4 mM CaCl2, and 0.4 mM MgCl2, which was adjusted to pH 7.2 with NaOH. The cells (2 ml) were inoculated into 1 liter of the same medium and then cultivated at 70°C. Cells were collected after 680 min, and then, the expression of each mRNA was analyzed.
为解析嗜热栖热菌HB8(Thermus thermophilus HB8)TTHA0344蛋白的功能,我们对在丰富培养基中于70℃培养680分钟的TTHA0344缺陷菌株的总信使核糖核酸(total mRNA)表达谱进行了分析。实验整体设计如下:将3株TTHA0344缺陷型嗜热栖热菌HB8菌株分别接种于3 mL TT培养基中,该培养基含0.8%蛋白胨(polypeptone)、0.4%酵母提取物(yeast extract)、0.2%氯化钠(NaCl)、0.4 mM氯化钙(CaCl₂)、0.4 mM氯化镁(MgCl₂),以氢氧化钠(NaOH)调节pH至7.2,随后于70℃预培养16小时。取2 mL菌液接种至1 L相同培养基中,于70℃继续培养;培养680分钟后收集菌体,对各mRNA的表达水平进行分析。
