A systematic investigation into the reactions of Cd(NO<sub>3</sub>)<sub>2</sub>·4H<sub>2</sub>O with o- and p-phenylenediacetic acid within the presence of the semi-rigid 1,4-bis(imidazol-1-ylmethyl)benzene (bimb) co-ligands under similar conditions was carried out. In this study, two novel coordination polymers containing Cd(II) ions as the metal ions {[Cd(μ-opda)(bimb)]·DMF}<sub>n</sub> (<b>1</b>, opda = 1,2-phenylenediacetate) and [Cd(μ-ppda)(bimb)<sub>0.5</sub>]<sub>n</sub> (<b>2</b>, ppda = 1,4-phenylenediacetate) have been prepared and structurally characterized. Furthermore, the inhibitory function of these two synthetic complexes on MG-63 human osteogenic sarcoma cells was evaluated with cell counting kit-8. And the Annexin V-FITC/PI apoptosis detection was performed to detect the induction ability of the complexes on the cancer cell apoptosis. Then, the western blot was performed to measure the activation level of the PTEN-AKT pathway.
本研究在相似条件下,以半刚性配体1,4-二(咪唑-1-基甲基)苯(bimb)为辅助配体,系统考察了四水合硝酸镉[Cd(NO₃)₂·4H₂O]分别与邻苯二乙酸、对苯二乙酸的反应。本研究成功制备并结构表征了两例以镉(II)为金属离子的新型配位聚合物:{[Cd(μ-opda)(bimb)]·DMF}<sub>n</sub>(编号1,opda为1,2-苯二乙酸根)与[Cd(μ-ppda)(bimb)<sub>0.5</sub>]<sub>n</sub>(编号2,ppda为1,4-苯二乙酸根)。此外,本研究采用细胞计数试剂盒-8(CCK-8)评估了这两种合成配合物对人成骨肉瘤MG-63细胞的抑制活性;通过Annexin V-FITC/PI凋亡检测法分析了配合物诱导癌细胞凋亡的能力;并采用蛋白质印迹法(Western Blot)检测了PTEN-AKT信号通路的激活水平。
