"Monocytes were isolated from bone marrow of male and female mice using magnetic-activated negative selection. Isolated monocytes were stimulated for 8 h in vitro with 1000 EVs/cell of E. histolytica A1 (EhA1) or E. histolytica B2 (EhB2) EVs or corresponding mock controls. After stimulation, monocyte RNA was isolated and immune responses to EV stimulation assessed using bulk RNA sequencing."
本研究采用磁激活阴性分选法,从雄性与雌性小鼠的骨髓中分离单核细胞(monocytes)。将分离得到的单核细胞置于体外环境中,以每细胞1000个细胞外囊泡(extracellular vesicles, EVs)的剂量,分别使用溶组织内阿米巴A1株(E. histolytica A1, EhA1)、溶组织内阿米巴B2株(E. histolytica B2, EhB2)的细胞外囊泡,或相应的模拟对照进行刺激,刺激时长为8小时。刺激结束后,提取单核细胞的总RNA,并通过批量RNA测序(bulk RNA sequencing)评估细胞外囊泡刺激所引发的免疫应答。
