To identify downstream transcription factors induced by retinoic acid, we stimulated SFZ cells with 10 μM retinoic acid for 24 hours and performed microarray analysis. Superficial zone cells of articular cartilage (SFZ) were prepared from femur and tibial joint of postnatal 4days ICR mice. SFZ cells were cultured with or without 10 μM retinoic acid for 24 hours, and subsequent isolation of total RNA for Affymetrix Mouse Genome 430 2.0 array.
为鉴定视黄酸(retinoic acid)诱导的下游转录因子,我们以10 μM视黄酸处理SFZ细胞24小时并进行微阵列分析。本研究中的SFZ细胞即关节软骨表层区细胞(Superficial zone cells of articular cartilage, SFZ),取自出生后4日的ICR小鼠股骨与胫骨关节。将SFZ细胞分别在含有与不含10 μM视黄酸的培养基中培养24小时后,提取总RNA用于Affymetrix小鼠基因组430 2.0微阵列分析。
