In order to perform transcriptional analyses of two stromal subsets carrying the gain-of-function mutation of CXCR4, bulk RNA Sequencing (RNA-Seq) was conducted on SSC and OPC sorted from WT (Cxcr4+/+) , heterozygous (Cxcr4+/1013) and homozygous (Cxcr41013/1013) mutant mice for Cxcr4. 为对两种携带CXCR4功能获得性突变的基质细胞亚群开展转录组分析,研究人员对来自Cxcr4基因野生型(Cxcr4+/+)、杂合突变型(Cxcr4+/1013)及纯合突变型(Cxcr41013/1013)小鼠的分选所得SSC与少突胶质前体细胞(oligodendrocyte progenitor cells, OPC)进行了批量RNA测序(RNA-Seq)。实验整体设计:基于上述不同Cxcr4基因型小鼠的SSC或OPC的RNA-seq数据,开展基因表达谱的比较分析。
