To investigate the effect of chronic continuous hypoxia (11%) oxygen on lifespan and neurological function in Ercc1 ?/- mice. We performed bulk RNA-seq of the cerebellum from mice that were Ercc1 ?/- normoxia, Ercc1 ?/- hypoxia, wildtype normoxia, wildtype hypoxia. Overall design: Differential gene expression analysis of cerebellar bulk RNA-seq from the 4 groups above, taken at the time when the normoxic mutant of a birth cohort reached euthanasia criteria, ages 15-19 weeks.
为探究11%氧浓度的慢性持续性低氧对Ercc1 ?/-小鼠寿命与神经功能的影响,我们对四组小鼠的小脑组织开展了批量RNA测序(bulk RNA-seq),四组分别为:Ercc1 ?/-常氧组、Ercc1 ?/-低氧组、野生型常氧组及野生型低氧组。
实验设计概要:针对上述四组小鼠的小脑批量RNA测序数据进行差异基因表达分析,采样时机为同出生队列的常氧组突变小鼠达到安乐死标准时,受试小鼠周龄为15至19周。
