Potencies of almorexant, SB-408124 and SB-334867 antagonists in inhibition of [3H]almorexant binding to the membrane preparations from HEK293 cells transiently expressing rHCRTR1 and rHCRTR2.

[3H]almorexant was used at a concentration equal to its Kd values of 3.4 nM and 0.5 nM at rHcrtR1 and rHcrtR2, respectively, in these competition binding experiments. Ki values for [3H]almorexant binding inhibition by various antagonists were calculated as described under “Materials and Methods”. Values are ± SEM of the Ki calculated from three independent experiments, each performed in duplicate.
