PMT Tropane alkaloid

We downloaded all orthologous sequences of TAa and TPSs of <i>D. stramonium</i>, <i>D. metel</i> and 11 species of Solanaceae to construct the orthogrpups. Downloades sequenceso of proteins and ORF were extracted using TransDecoder and corroborated with GenScan. TPS genes were translated with GenScan, are in the right direction and are the correct CDS; we corroborated this with the NCBI. The database used for BLAST was Uniprot; all available data for the analyzed plant species were downloaded. These amounts 847,544 3. BLAST was carried out with a filtering evalue of 1e-³, and only hits above 85 % were selected.

我们下载了曼陀罗（*D. stramonium*）、洋金花（*D. metel*）以及11种茄科（Solanaceae）植物的TAa与TPSs直系同源序列，以构建直系同源基因家族（orthogroups）。利用TransDecoder工具提取下载得到的蛋白质序列与开放阅读框（ORF），并通过GenScan基因预测软件进行验证。经GenScan翻译得到的TPS基因，其序列方向与编码序列（CDS）均正确，我们已通过美国国家生物技术信息中心（NCBI）完成了该结果的验证。本次BLAST分析采用的数据库为UniProt数据库，我们下载了所分析植物物种的全部可用数据，共计847,544条。BLAST分析设置的过滤E值为1e-3，仅保留序列相似度≥85%的命中结果。
