CD74 activation was previously shown to affect gene transcription. The goal of this study was to research its role as a transcriptor regulator. CD74 was activated in primary CLL cells obtained from patients for 1 h. The cells were then fixed using DSG and formaldhyde, and ChIP-seq was conducted. Overall design: ChIP-seq was done for CD74 activated or non activated CLL cells with Ab for CD74 (4 replicates), CD74 activated H3K4Me2 (2 replicates), CD74 non-activated H3K4Me2 (2 replicates), H3K4Me3 (2 replicates), H3K27Ac (2 replicates).
此前已有研究证实,CD74的激活可影响基因转录。本研究旨在探究CD74作为转录调控因子的功能。实验中将从患者体内分离的原发性慢性淋巴细胞白血病(Chronic Lymphocytic Leukemia, CLL)细胞进行1小时的CD74激活处理,随后使用丁二酰亚胺戊二酸酯(DSG)与甲醛对细胞进行固定,并开展染色质免疫共沉淀测序(ChIP-seq)实验。整体实验设计如下:针对CD74激活与未激活的CLL细胞,使用对应抗体完成ChIP-seq检测,具体分组及重复次数为:靶向CD74的实验组(4次生物学重复)、CD74激活状态下的H3K4Me2组(2次生物学重复)、CD74未激活状态下的H3K4Me2组(2次生物学重复)、H3K4Me3组(2次生物学重复)以及H3K27Ac组(2次生物学重复)。
