We cultured D. immitis L3 and L4 of two laboratorial strains with different susceptibility status to macrocyclic lactone drugs in vitro. Excretory/secretory microRNAs were sequenced and analyzed. Overall design: D. immitis L3 and L4 of two different strains were allowed to excrete/secrete miRNAs in vitro, which were profiled by miRNA sequencing and compared.
本研究体外培养了两株对大环内酯类药物(macrocyclic lactone drugs)易感性存在差异的实验室犬恶丝虫(Dirofilaria immitis,缩写D. immitis)菌株的第三期幼虫(L3)与第四期幼虫(L4),并对其排泄/分泌型微小RNA(microRNA, miRNA)开展了测序与分析。整体实验设计:将两株不同菌株的犬恶丝虫L3与L4置于体外环境中培养以收集其排泄/分泌的miRNA,通过miRNA测序构建miRNA表达谱并进行组间比较。
