We show that numerous miRNAs are transcriptionally up-regulated in papillary thyroid carcinoma (PTC) tumors compared with unaffected thyroid tissue. Among the predicted target genes of the three most upregulated miRNAs (miRs 221, 222 and 146b), only less than 15% showed significant downexpression in transcript level between tumor and unaffected tissue. The KIT gene which is known to be downregulated by miRNAs 221 and 222 displayed dramatic loss of transcript and protein in those tumors that had abundant mir-221, mir-222, and mir-146b transcript. Keywords: Disease state analysis Total RNA was extracted from paired tumor and normal thyroid tissues from 9 PTC patients. The same set samples were applied to Custom miRNA microarray chips (OSU_CCC version 2.0) and Affymetrix HG-U133 plus 2 chips.
本研究证实,相较于正常甲状腺组织,甲状腺乳头状癌(papillary thyroid carcinoma, PTC)肿瘤内大量微小RNA(microRNA, miRNAs)呈现转录水平上调。在这三个上调幅度最高的miRNA(miR-221、miR-222与miR-146b)的预测靶基因中,仅不足15%的基因在肿瘤组织与正常甲状腺组织间的转录水平呈现显著下调。已知可被miR-221与miR-222靶向下调的KIT基因,在miR-221、miR-222及miR-146b表达丰度较高的肿瘤中,其转录本与蛋白水平均出现显著降低。
