To determine effects of p53 activation on steady-state mRNA levels, we performed RNA-seq analysis of colorectal carcinoma cell line HCT116 (p53+/+ and p53 -/-), breast carcinoma line MCF7, and osteosarcoma line SJSA treated with MDM2 inhibitor Nutlin. Overall design: Total RNA was sequenced from HCT116 (p53+/+ and p53 -/-), MCF7 and SJSA cells treated with DMSO or 10 microM Nutlin for 12 hours.
为探究p53激活对细胞稳态mRNA水平的影响,我们对经MDM2抑制剂Nutlin处理的结直肠癌细胞系HCT116(p53+/+与p53-/-)、乳腺癌细胞系MCF7以及骨肉瘤细胞系SJSA开展了RNA测序(RNA-seq)分析。
实验整体设计:提取经二甲基亚砜(DMSO)或10 μM Nutlin处理12小时的HCT116(p53+/+与p53-/-)、MCF7及SJSA细胞的总RNA,随后进行测序。
