Zebrafish of the Nadia strain were raised at 28.5°C, 34°C, or 36°C from 18-30 days post-fertilization (dpf) to understand the developmental effects of high temperature on zebrafish that possess a genetic sex determinant. Fish were evaluated for genotypic and phenotypic sex and RNA-seq was performed on gonads collected at 90dpf. Strand-specific sequencing libraries were prepared with the BIOO NEXTflex Rapid Directional qRNA-Seq kit and sequenced on an Illumina HiSeq 4000.

为探究高温对携带遗传性别决定因子的斑马鱼的发育影响,本研究将Nadia品系斑马鱼于受精后18~30天(days post-fertilization,简称dpf)期间,分别置于28.5°C、34°C及36°C环境中饲养。后续对实验个体开展基因型性别与表型性别鉴定,并于受精后90天(90dpf)采集性腺开展RNA测序(RNA-seq)。实验采用BIOO NEXTflex Rapid Directional qRNA-Seq试剂盒构建链特异性测序文库,并在Illumina HiSeq 4000测序平台上完成测序。
