In order to understand LBG derived galacto-manno-oligosaccharides utilization by a probiotic bacterium, Lactobacillus plantarum WCFS1, we have grown Lactobacillus plantarum WCFS1 (in duplicates) till mid log phase (OD600nm ~0.5, 10 h) in carbon free MRS (de Man, Rogosa Sharpe ) media containing either galacto-manno-oligosaccharides, mannose, glucose or galactose (1% w/v) as the sole carbon source. 为解析益生菌植物乳杆菌(Lactobacillus plantarum)WCFS1对LBG衍生的半乳甘露寡糖(galacto-manno-oligosaccharides)的利用机制,我们将植物乳杆菌WCFS1设置两组生物学重复,在以1%(w/v)半乳甘露寡糖、甘露糖、葡萄糖或半乳糖为唯一碳源的无碳源MRS(de Man, Rogosa Sharpe)培养基中培养至对数生长期中期(OD600nm≈0.5,培养时长10 h)。总体实验设计:实验菌株为植物乳杆菌(Lactobacillus plantarum);采用安捷伦(Agilent)乳杆菌基因表达8×15k芯片(阵列编号AMADID: 067475),该芯片由吉纳迪克技术私人有限公司(Genotypic Technology private Limited)定制。
