We analyzed function of ERRα in human skin fibroblasts which silenced ERRα gene expression by microarray analysis. TIG113 was seeded in a culture dish, cultured until subconfluent, and then transfected with siRNA. After 48 h, total RNA was extracted from siRNA transfected or control TIG113 cells.
我们通过微阵列分析(microarray analysis)沉默雌激素相关受体α(ERRα)的基因表达,以此探究其在人类皮肤成纤维细胞中的功能。具体实验流程如下:将TIG113细胞接种于培养皿中,培养至亚汇合状态,随后以小干扰RNA(siRNA)进行转染。转染48小时后,分别从经siRNA转染的TIG113细胞及对照组TIG113细胞中提取总RNA。
