Using mass spectrometry-based label-free quantitative (LFQ) proteomics analysis of in vitro differentiated murine Th17 and induced T regulatory (iTreg) cells. More than 4000 proteins covering almost all subcellular compartments were detected. Quantitative comparison of the protein expression profiles resulted in the identification of proteins specifically expressed in the Th17 and iTreg cells. Importantly, our combined analysis of proteome and gene expression data revealed protein expression changes that were not associated with changes at the transcriptional level. Overall design: Naïve CD4+ T cells were isolated from 8–10-week-old C57BL/6 mice spleens. Cells were polarized to Th0, Th17 and iTreg conditions for three days. Four biological replicates were prepared for RNA-seq.
本数据集采用基于质谱的无标记定量(label-free quantitative, LFQ)蛋白质组学分析方法,对体外分化的小鼠Th17细胞与诱导性调节性T(induced T regulatory, iTreg)细胞进行检测分析。本次研究共检测到超过4000种蛋白质,覆盖几乎所有亚细胞区室。通过对蛋白质表达谱进行定量比较,成功鉴定出仅在Th17细胞与iTreg细胞中特异性表达的蛋白质。值得注意的是,我们对蛋白质组与基因表达数据进行整合分析后发现,部分蛋白质的表达变化并未伴随转录水平的改变。整体实验设计如下:从8~10周龄C57BL/6小鼠脾脏中分离初始CD4+ T细胞,将其分别置于Th0、Th17及iTreg细胞极化培养条件下培养三天。本次实验设置4个生物学重复样本用于RNA测序(RNA-seq)。
