To identify the differentially expressed genes in normal gastric mucosa and gastric cancer tissues, we employed the microarray profiling analysis. Genes with greater than 2-fold change and P-value <0.05 were identified as differentially expressed genes. From this dataset, we obtained differentially expressed genes by cluster analysis and enriched GOs was identified by Gene set enrichment analysis (GSEA) analysis, microtubule GO was one of the most enriched GOs, and the gene KIF14 was selected to go on to the next step in the study. Overall design: 10 paired of total samples were analyzed. Genes with an FDR 0.25 and a fold-change ≥2 were selected. Pathway and GO analysis was used for the next step analysis.
为鉴定正常胃黏膜与胃癌组织中的差异表达基因(differentially expressed genes),本研究采用芯片表达谱分析(microarray profiling analysis)技术。以表达变化倍数大于2倍且P值<0.05的基因作为差异表达基因的筛选阈值。本数据集通过聚类分析获取差异表达基因,并通过基因集富集分析(Gene Set Enrichment Analysis, GSEA)鉴定富集的基因本体(Gene Ontology, GO)条目,其中微管相关GO条目为显著富集的GO条目之一,后续研究选取基因KIF14开展下一步实验。
整体实验设计:共分析10对配对样本。筛选标准为错误发现率(False Discovery Rate, FDR)≤0.25且表达变化倍数≥2的基因。后续分析采用通路富集分析与GO富集分析方法。
