This study was based on a panel of 180 common bean (P. vulgaris) genotypes representative of the genetic diversity of a common bean germplasm repository of 1,800 accessions, deposited at the Agronomic Institute (IAC) in Campinas, Brazil. The IAC was founded in 1887, and is an important research institute of São Paulo state’s Department of Agriculture and Supply.Two GBS libraries were constructed in 95-plex (190 accessions). Genomic DNA was co-digested with the restriction enzyme ApeKI and barcoded adapters were ligated to individual samples. Each library was single-end sequenced in a single lane of Genome Analyzer II (Illumina, Inc.). These procedures were carried out at the Genomic Diversity Facility, Institute of Biotechnology, Cornell University, USA.

本研究选取巴西坎皮纳斯市农业研究所(Agronomic Institute, IAC)所保存的1800份普通菜豆种质资源库中具有代表性的180份普通菜豆(Phaseolus vulgaris, P. vulgaris)基因型作为试验群体。该研究所(IAC)始建于1887年,是圣保罗州农业与供应部下属的重要科研机构。 本研究构建了2个95重的基因组分型测序(Genotyping-by-Sequencing, GBS)文库,共涉及190份种质材料。实验过程中,先使用限制性内切酶ApeKI对基因组DNA进行酶切,随后为每个独立样本连接带条形码的接头(barcoded adapters);每个文库均在Illumina公司的Genome Analyzer II测序仪的单个泳道上完成单端测序。上述实验流程均在美国康奈尔大学生物技术研究所基因组多样性中心(Genomic Diversity Facility, Institute of Biotechnology, Cornell University)完成。
