Mice were fed Se-deficient or Se-adequate diets for 6 weeks. Liver and lung tissue were harvested and processed for RNA-Seq, ribosome profiling, and microarray analysis. From these studies, we identified changes in mRNA levels and translation of selenoprotein genes and genes regulated by interferon-gamma. Cytokine profiles of serum indicated that interferon-gamma and IL-6 levels were increased in the Se-adequate mice relative to Se-deficient mice. 本研究将小鼠分为两组,分别饲喂硒缺乏(Se-deficient)饲料与硒充足(Se-adequate)饲料,持续6周。随后采集两组小鼠的肝脏与肺组织,开展RNA测序(RNA-Seq)、核糖体谱分析(ribosome profiling)及微阵列(microarray)分析。通过上述实验,我们鉴定出硒蛋白基因以及受干扰素-γ(interferon-gamma)调控的基因的mRNA水平与翻译过程的变化。血清细胞因子谱分析结果显示,相较于硒缺乏组小鼠,硒充足组小鼠血清中的干扰素-γ与白细胞介素-6(IL-6)水平显著升高。整体实验设计:对饲喂硒缺乏或硒充足饲料的小鼠的肝脏组织进行RNA测序(RNA-Seq)分析。
