To better understand how T21 affects N-glycosylation during neural differentiation, we comprehensively characterized and compared the total N-glycomes of induced pluripotent stem cells (iPSCs) and their neural stem cell (NSC) derivatives. We utilized an isogenic in vitro model of T21 in which both T21 and euploid disomic karyotype (D21) clones were obtained from a single individual with mosaic DS. N-glycomics analysis of whole cell lysates was performed using liquid chromatography coupled with tandem mass spectrometry to determine proposed N-glycan structures. Here we upload the raw data files for the analysis.

为更清晰地阐明三体21(T21)在神经分化过程中对N-糖基化的影响,我们对诱导多能干细胞(induced pluripotent stem cells,iPSCs)及其分化所得的神经干细胞(neural stem cell,NSCs)的总N-糖组进行了全面表征与对比分析。本研究采用同基因体外T21模型,该模型的三体21与整倍体二倍体核型(D21)克隆均来源于同一嵌合型唐氏综合征个体。我们通过液相色谱-串联质谱法对全细胞裂解物开展N-糖组学分析,以推定N-聚糖的具体结构。本文上传本次分析所用的全部原始数据文件。
