The NanoString analysis of 770 genes expressed by 24 immune cell types in the tumors at 10 days after injection were performed. The Nanostring analysis uncovered marked difference in key immune regulatory networks associated with T, DC, NC and macrophage function Yumm1.5 tumor cells were injected into WT or Rnf5-/- mice. Tumor were collected at day 10 and RNA were isolated from the tumor. Then Nanostring nCounter assay for immune genes were performed
本研究针对接种后10天的肿瘤组织中24种免疫细胞类型所表达的770个基因开展了NanoString(纳米串)分析。该分析揭示,与T细胞、树突状细胞(Dendritic Cell, DC)、NC及巨噬细胞功能相关的关键免疫调控网络存在显著差异。本实验将Yumm1.5肿瘤细胞接种至野生型(Wild Type, WT)或Rnf5基因敲除(Rnf5-/-)小鼠体内,于接种后第10天收集肿瘤组织并从中提取总RNA,随后针对免疫基因完成了NanoString nCounter检测。
