A2059G mutation on 23s rDNA mutation was responsible for high level azithromycin resistance in Neisseria gonorrhoeae. Four copies 23s rDNA A2059G mutation in high level azithromycin resistance ZJXSH89 clinical isolate and four copies 23s rDNA G2059A mutation in azithromycin sensitive ZJXSH86 clinical isolate had been made. The genome sequences of the wild type and mutant were compared to reveal whether secondary mutation exist or not.
23S核糖体DNA(23S rDNA)的A2059G突变可导致淋病奈瑟菌(Neisseria gonorrhoeae)产生高水平阿奇霉素(azithromycin)耐药性。本研究构建了携带4个拷贝23S rDNA A2059G突变的高水平阿奇霉素耐药临床分离株ZJXSH89,以及携带4个拷贝23S rDNA G2059A突变的阿奇霉素敏感临床分离株ZJXSH86。通过比对野生型菌株与突变株的基因组序列,以明确是否存在次级突变(secondary mutation)。
