Function of Ets1 in regulating macrophage polarization and adipose inflammation

To investigate the cooperative function Ets1 in macrophage, we generats Ets1 knockout mice.We then performed gene expression profiling analysis in macrophages seperated from either control or ko mice. We also performed Ets1 ChIP-seq, using M1 and M2 macrophages seperated from wild type C57 bone marrow, for mechanism study. Transcription sequencing (RNA-seq) in macrophages seperated from either Ets1 null or control mice, were perfomed. Chromatin immunoprecipitation DNA-sequencing (ChIP-seq) for Ets1 in M1 and M2 macrophages, were perfomed.

为探究Ets1在巨噬细胞（macrophage）中的协同功能，我们构建了Ets1基因敲除小鼠。随后，我们对来自对照小鼠与Ets1敲除小鼠的巨噬细胞开展了基因表达谱分析。为开展机制研究，我们利用从野生型C57小鼠骨髓中分离得到的M1型与M2型巨噬细胞，进行了Ets1的染色质免疫共沉淀测序（Chromatin Immunoprecipitation sequencing, ChIP-seq）。我们还对来自Ets1基因缺失型与对照小鼠的巨噬细胞完成了转录测序（RNA-seq）。此外，我们针对M1与M2型巨噬细胞中的Ets1开展了染色质免疫共沉淀测序（ChIP-seq）实验。
