The expression of the forkhead transcription factor CHES1, also known as FOXN3, is reduced in many types of cancers. In vitro, CHES1 expression suppresses cell proliferation in tumor cell lines but not in normal cells. Conversely shRNA-mediated depletion of CHES1 increases tumor cell proliferation. We used microarrays to characterize the gene expression changes induced by CHES1 in H1299 cancer cells Overall design: H1299 cells were infected with a retroviral vector that expresses CHES1 or an empty vector. After 5 days of infection, RNA was extracted from three independent replicas of each condition. Total RNA was send to Genome Quebec service for hybridization with Affymetrix microarrays.

叉头转录因子CHES1(又称为FOXN3)的表达在多种癌症类型中均出现下调。体外实验表明,CHES1的表达可抑制肿瘤细胞系的增殖,但对正常细胞无此抑制效应。与之相反,通过短发夹RNA(short hairpin RNA,shRNA)介导的CHES1敲低,会促进肿瘤细胞增殖。本研究采用微阵列技术,对H1299癌细胞中CHES1诱导的基因表达变化进行表征。实验总体设计:将H1299细胞分别感染携带CHES1编码序列的逆转录病毒载体与空载对照载体。感染5天后,从两种实验条件的三个独立生物学重复样本中提取总RNA,随后将总RNA送至魁北克基因组(Genome Quebec)服务平台,开展Affymetrix微阵列杂交检测。
