Aim: The goals of this study are to compare transcriptome profile of mESCs and differentiated cells, and transcriptome profile of brains from wild type and Discn-/- mice. Overall design: mESCs and differentiated cells derived from mESCs were treated with 10 µM etoposide or 2 mM Hydroxyurea for 4 hours. Brains were isolated from 5 weeks old wild type (WT) and Discn-/- mice. RNA was isolated and RNA-seq was performed.

研究目的:本研究旨在对比小鼠胚胎干细胞(mouse embryonic stem cells,mESCs)与分化细胞的转录组谱,以及野生型(wild type,WT)与Discn-/-小鼠脑部的转录组谱。实验设计概述:将小鼠胚胎干细胞(mESCs)及其分化所得细胞分别用10 μM依托泊苷(etoposide)或2 mM羟基脲(Hydroxyurea)处理4小时。从5周龄野生型(WT)及Discn-/-小鼠体内分离脑组织。提取总RNA并进行RNA测序(RNA-seq)。
