本研究通过RNA干扰(RNAi)技术在人类神经母细胞瘤细胞系中敲低MYCN基因。MYCN基因敲低可引发全基因组表观组改变,具体包括关键DNA修复基因区域的核小体富集,以及启动子区域H3K9乙酰化(H3K9ac)水平下调。本研究针对对照组(ctrl)与MYCN敲低组(N-myc_kd)的神经母细胞瘤细胞系BE2C,开展了6种组蛋白修饰的染色质免疫沉淀测序(ChIP-seq)检测。
