Immunodeficient male BALBc nude mice were obtained from the Chongqing Pharscin InnoBio Experimental Animal Center. HCT116 CRC cells were subcutaneously injected into the right flank of each mouse. When palpable tumors had developed (day 7), the mice were randomly assigned to two groups (n = 4 per group) and administered intraperitoneal injections of Epimedium sagittatum derived extracellular vesicles (ESEVs) (0 or 22 mg/kg per mouse) once daily for 14 consecutive days. At the end of treatment (day 21), all mice were euthanized; tumors were excised and weighed. Tumor sample was used for transcriptomic analysis. To clarify molecular mechanisms underlying the antitumor effects of ESEVs, RNA sequencing was performed on colorectal tumor tissues from mice treated with ESEVs or NaCl (control). Eight cDNA libraries were generated, comprising four biological replicates per group.
免疫缺陷雄性BALBc裸小鼠购自重庆菲斯克因诺生物(Pharscin InnoBio)实验动物中心。将HCT116结直肠癌细胞(Colorectal Cancer, CRC)皮下接种于每只小鼠的右侧背部。待可触及肿瘤形成(第7天)时,将小鼠随机分为两组(每组n=4),分别腹腔注射淫羊藿来源细胞外囊泡(Epimedium sagittatum derived extracellular vesicles, ESEVs),给药剂量为每只小鼠0或22 mg/kg,每日一次,连续给药14天。给药结束后(第21天),处死所有小鼠,剥离肿瘤并称重。取肿瘤样本用于转录组学分析。为阐明ESEVs抗肿瘤作用的潜在分子机制,对接受ESEVs或氯化钠(NaCl,对照组)处理的小鼠结直肠肿瘤组织开展RNA测序。本次实验共构建8个cDNA文库,每组包含4个生物学重复。
