To identify the target genes of Evi-1 in hematopoietic stem cells (HSCs), we carried out genome-wide transcriptional analysis using wild-type and Evi-1-deleted HSCs. Overall design: Lineage(-), Sca-1(+), c-Kit(+) cells derived from Evi-1(flox/flox) mice were transduced with GFP or Cre-GFP expressing retroviruses. GFP(+) cells were sorted and and analyzed by Affymetrix® Mouse Genome 430 2.0 Array® for gene expression. Two independent experiments were performed.

为鉴定造血干细胞(hematopoietic stem cells, HSCs)中Evi-1的靶基因,我们采用野生型与Evi-1缺失的造血干细胞开展了全基因组转录组分析。实验整体设计:将源自Evi-1(flox/flox)小鼠的谱系阴性(Lineage(-))、Sca-1阳性(Sca-1(+))、c-Kit阳性(c-Kit(+))细胞,使用表达绿色荧光蛋白(Green Fluorescent Protein, GFP)或Cre-GFP的逆转录病毒进行转导。随后分选GFP阳性细胞,通过Affymetrix® Mouse Genome 430 2.0 Array®开展基因表达分析。本研究共完成两次独立实验。
