The purpose of this study was to characterize the microbial functional and structural diversity with depth across representative redox zones of a hydrocarbon plume, and an adjacent uncontaminated wetland, at the Bemidji Oil Spill site. A combination of quantitative PCR, denaturing gradient gel electrophoresis, and pyrosequencing were applied to vertically sampled sediment cores.
本研究旨在表征贝米吉石油泄漏场地(Bemidji Oil Spill site)内,烃类羽流(hydrocarbon plume)的典型氧化还原带及其邻近未受污染湿地中,微生物功能与结构多样性随深度的分布特征。研究采用定量聚合酶链式反应(quantitative PCR)、变性梯度凝胶电泳(denaturing gradient gel electrophoresis)与焦磷酸测序(pyrosequencing)相结合的方法,对垂直采集的沉积物岩芯进行分析。
