Anti CD90 was used to deplete ILCs in RAG mice at point of iniation of DSS colitis in Rag mice. After 6 days the Macrophages were sorted from the colon and compared by using RNASeq to mice in which ILCs had not been repleted but still had colitus induced. All mice recived were RAG KO mice and reciveed DSS. 6 recived anti CD90 and the remaining 6 recived an isotype control
本实验所用小鼠均为RAG基因敲除小鼠(RAG KO mice),且全部接受了葡聚糖硫酸钠(Dextran Sulfate Sodium, DSS)结肠炎造模。在DSS结肠炎造模的起始阶段,6只小鼠经抗CD90抗体(Anti CD90)处理以耗竭体内的固有淋巴细胞(Innate Lymphoid Cells, ILCs),剩余6只则给予同型对照抗体(isotype control)处理。造模6天后,从两组小鼠的结肠中分选巨噬细胞(Macrophages),通过RNA测序(RNASeq)对两组样本进行转录组比较分析:一组为经抗CD90抗体处理后耗竭了ILCs的小鼠,另一组则未耗竭ILCs但同样诱导了结肠炎。
