Gene expression profile at single cell level of lung and tracheobronchial lymph nodes leukocytes of healthy and influenza-infected pigs

We characterized the single cell landscape of lung leukocytes of healthy pigs and then compared them to pigs infected with H1N1 A/California/04/2009 with or without oseltamivir antiviral therapy. Live lung leukocytes of 6-week-old mixed breed pigs were isolated by fluorescence-activated cell sorting (FACS) according to the absence of propidium iodide signal and analyzed using scRNAseq.

本研究首先解析了健康猪肺白细胞（lung leukocytes）的单细胞图谱，随后将其与感染甲型H1N1流感病毒A/California/04/2009（H1N1 A/California/04/2009）、分别接受或不接受奥司他韦（oseltamivir）抗病毒治疗的感染猪肺白细胞进行对照分析。本研究通过荧光激活细胞分选术（fluorescence-activated cell sorting, FACS），依据碘化丙啶（propidium iodide）信号阴性筛选出6周龄混合品种猪的活肺白细胞，并采用单细胞RNA测序（scRNAseq）对其进行分析。
