In this experiment, we evaluted whole transcriptomic changes in ex vivo cultured ducts expressing mutant Kras compared to control ducts. The cells were cultured for 7 days before RNA was isolated for RNA-seq. For the ex vivo cultured ducts, Hnf1b:CreERT2; LSL-KrasG12V mice were given 10 mg tamoxifen or corn oil control and euthanized at day 4 for ex vivo culturing of ducts.

本实验中,我们对比了表达突变型KRAS的离体(ex vivo)培养导管与对照导管的全转录组变化。实验前将导管相关细胞培养7天,随后提取RNA进行RNA测序(RNA-seq)。针对用于离体培养的导管,我们向Hnf1b:CreERT2; LSL-KrasG12V 小鼠注射10 mg他莫昔芬或玉米油作为对照,并于给药后第4天对小鼠实施安乐处死,以获取导管用于后续离体培养。
