We randomly picked two male and two female shrimps of Fenneropenaeus chinensis at 5-months-old. Muscle, gonad, hepatopancreas, intestine, ganglion, heart, and sputum were collected and extracted total RNA using Trizol (Invitrogen, USA). Twenty-eight RNA samples (4 individuals×7 tissues) were mixed into one pool with equal amount of nucleic acids. The mixed pool was used for single-molecule full-length transcriptome sequencing by PacBio platform.Another 15 female and 15 male shrimps were chosen to perform Illumina-based RNA-seq. We collected their gonad and muscle and extracted total RNA using Trizol (Invitrogen, USA). Each 5 RNA samples of same sex and same tissue were mixed into one pool. The 3 pools of same tissue and sex were treated as biological duplicates.

我们随机选取4只5月龄的中国明对虾(Fenneropenaeus chinensis),雌雄各2只。采集其肌肉、性腺、肝胰腺、肠道、神经节、心脏及鳃组织,采用Trizol试剂(Invitrogen,美国)提取总RNA。共获得28份RNA样本(4个个体×7种组织),按照等摩尔核酸含量混合为一个混合池。该混合池用于PacBio平台的单分子全长转录组测序。另外选取15只雌性与15只雄性中国明对虾,开展基于Illumina平台的RNA测序(RNA-seq)。采集其性腺与肌肉组织,同样采用Trizol试剂(Invitrogen,美国)提取总RNA。将同性别、同组织的5份RNA样本混合为一个混合池,每类同性别同组织的3个混合池作为生物学重复。
