To learn more about the role of FRUITFULL (FUL), in pistil/silique development, we performed a ChIP-seq experiment to identify direct targets of FUL in the pistil/silique. Overall design: We used a pFUL:FUL-GFP line to conduct chromatin immuno-precipitation experiments with GFP-specific antibodies followed by deep-sequencing (ChIP-seq) in order to determine FUL binding sites on a genome-wide scale. For the samples, pistils/siliques from flower stages 12-16 were harvested, fixed for 15-30’ and the immunoprecipitation was performed according to Kaufmann et al., 2010 (doi: 10.1038/ nprot.2009.244.). Experiments were done in two biological replicates with the according input samples as controls.

为深入解析FRUITFULL(简称FUL)在雌蕊/长角果发育中的调控功能,我们开展了染色质免疫共沉淀测序(ChIP-seq)实验,以鉴定FUL在雌蕊/长角果中的直接靶基因。整体实验设计:我们采用pFUL:FUL-GFP转基因株系,使用GFP特异性抗体进行染色质免疫共沉淀实验,随后进行深度测序(ChIP-seq),从而在全基因组范围内定位FUL的结合位点。实验材料为花发育阶段12至16期的雌蕊/长角果,样品固定时长为15~30分钟,免疫共沉淀实验严格遵循Kaufmann等人2010年发表的实验方案(DOI: 10.1038/nprot.2009.244)开展。本实验设置两组生物学重复,并以对应的输入样品作为对照。
