Cadmium is a widespread environmental heavy metal pollutant as well as a strong endocrine disruptor. Epidemiologic evidence shows a positive correlation between cadmium levels in humans and the incidence of diabetes mellitus.Moreover, cadmium can accumulate in pancreatic islet and induce oxidative damage, apoptosis and decreased insulin.However, the specific mechanisms involved have not been completely elucidated.We hope to use microarrays to explore the mechanism by analyzing the changes of genes during the process of cadmium damage in islet β-cells. Overall design: After treatment of NIT-1 cells with 4 μmol/L CdSO4 to induce oxidative damage, total RNA was collected from the experimental and control groups.
镉(Cadmium)是一种广泛分布的环境重金属污染物,同时也是强效内分泌干扰物。流行病学证据表明,人体内镉水平与糖尿病(diabetes mellitus)发病率呈正相关。此外,镉可在胰岛(pancreatic islet)中蓄积,并诱发氧化损伤、细胞凋亡及胰岛素分泌降低。然而,其所涉及的具体分子机制尚未完全阐明。本研究拟通过基因芯片(microarrays)分析胰岛β细胞在镉损伤过程中的基因表达变化,以探索其潜在作用机制。实验设计:使用4 μmol/L硫酸镉(CdSO4)处理NIT-1细胞以诱导氧化损伤后,分别收集实验组与对照组的总RNA。
