To investigate the whole fungal transcriptional changes induced by CtBOT6 in Colletotrichum tofieldiae 4 (Ct4), we performed RNA-seq analysis on CtBOT6 constitutive expression transformants (OX#1, OX#10, OX#24) compared to Ct4 WT. To obtain candidate genes regulated by CtBOT6, we employed in vitro growth conditions in which we can detect the synthesis of the botrydial precursors in the OX lines while excluding any potential influence from plants. Three-day-old mycelia cultured in liquid media were used for RNAseq analysis.
为探究菲氏炭疽菌4号菌株(Colletotrichum tofieldiae 4, Ct4)中CtBOT6诱导的全真菌转录组变化,我们针对CtBOT6组成型表达转化子(OX#1、OX#10、OX#24)与Ct4野生型(WT)开展了RNA测序(RNA-seq)分析。为获取受CtBOT6调控的候选基因,我们采用体外培养体系,该体系可在排除植物潜在干扰的前提下,检测到OX株系中葡萄孢素(botrydial)前体的合成。我们选取液体培养基中培养3天的菌丝体进行RNA测序分析。
