To investigate the mechanism underlying the altered localization of pigmented melanosomes in cells expressing activated beta-catenin, we extracted mRNA from WT and bcat* cell lines and performed microarray-based transcriptome analysis. We previously studied the effect of beta-catenin on the melanocyte lineage by creating a transgenic mouse model containing a stable, transcriptionally active beta-catenin mutant protein specifically in the melanocyte lineage (bcat* mice; Delmas et al., Genes Dev 2007). Melanocytes from the skin of wild-type C57BL/6J and bcat* mice were isolated and different cell lines were established in culture. In this study, we performed Affymetrix transcription microarray experiments on two wild-type (9v and 14d) and two bcat* (10d and 13d) cell lines.

为探究激活型β-连环蛋白(beta-catenin)表达细胞中色素化黑素体(pigmented melanosomes)定位异常的潜在机制,我们从野生型(Wild Type, WT)与bcat*细胞系中提取mRNA,并开展了基于微阵列(microarray)的转录组分析。 此前我们通过构建仅在黑素细胞谱系(melanocyte lineage)中表达稳定转录活性β-连环蛋白突变蛋白的转基因小鼠模型(bcat*小鼠;Delmas等,《Genes Dev》2007),研究了β-连环蛋白对黑素细胞谱系的调控效应。 我们分离了野生型C57BL/6J小鼠与bcat*小鼠皮肤来源的黑素细胞,并建立了多株体外培养细胞系。 本研究针对两株野生型细胞系(9v与14d)以及两株bcat*细胞系(10d与13d)开展了Affymetrix转录微阵列实验。
