The gene expression level of CTTN was decreased in HPV-negative HNSCC cell line with an amplification and overexpression of CTTN by performing knockdown experiments using RNA interference. Gene expression profiling and consecutive pathway analyses showed a number of deregulated genes known to be involved in metastatic processes such as migration and invasion. 本研究利用RNA干扰(RNA interference)技术开展基因敲低实验,结果表明,在携带CTTN扩增与过表达的人乳头瘤病毒(HPV)阴性头颈部鳞状细胞癌(head and neck squamous cell carcinoma,HNSCC)细胞系中,CTTN的基因表达水平显著下调。随后通过基因表达谱分析与相继通路分析,本研究鉴定出一批已知参与迁移、侵袭等肿瘤转移过程的失调基因。此外,本研究还明确了CTTN的三个潜在下游靶基因:MAF、MAFB与PLAC2。本实验设置CTTN敲低组与对照组,每组设置4个生物学重复。
