A laboratory strain of Ae. aegypti (Liverpool strain) was exposed to ibuprofen (1µg/L)from larvae hatching to imagoes emergence. To understand the mechanistic processes underlying the direct and intergenerational impacts of ibuprofen, we used RNA-seq based transcriptomic approach. Each condition (exposed or not to ibuprofen) was analyzed in triplicate, each RNA sample being extracted from 2-4 days old non-blood fed adult females (exposed or not) and from their progeny (L1 larvae exposed or not) . RNA-seq libraries were sequenced on an illumina GAIIx sequencer as 150 bp single reads.
本研究使用埃及伊蚊(Ae. aegypti)利物浦品系的实验室种群,在其从幼虫孵化至成虫羽化的整个生命周期中,均暴露于浓度为1μg/L的布洛芬环境中。为解析布洛芬直接影响及代际影响背后的分子机制,我们采用了基于RNA测序(RNA-seq)的转录组学分析方法。每种处理组(布洛芬暴露组与未暴露对照组)均设置三次生物学重复;每份RNA样本分别取自2~4日龄未吸血的成年雌蚊(暴露组或对照组)及其子代(暴露或未暴露的L1期幼虫)。RNA测序文库在Illumina GAIIx测序仪上以150bp单端读长模式完成测序。
