miRNA regulate gene expression at the post-transcriptionnal level. To gain further insight into this process, we analysed by Affymetrix microarray, the transcriptome of Dicer WT or Dicer deleted mouse CD4 T cells. Overall design: Mouse CD4 T cells were sorted by flow cytometry from control CD4Cre neg Dicer lox/lox or CD4Cre pos Dicer lox/lox animals, total RNA was extracted using RNA bee and amplified before hybridization onto Mouse Gene arrays. Three biological replicate were analysed for each condition (3 Dicer WT control and 3 Dicer KO).
微小RNA(microRNA,miRNA)可在转录后水平调控基因表达。为深入解析该调控过程,我们采用Affymetrix基因芯片(Affymetrix microarray)对Dicer野生型(Wild Type, WT)及Dicer敲除的小鼠CD4阳性T细胞的转录组开展了分析。实验设计概述:通过流式细胞术(flow cytometry),我们从对照CD4Cre阴性Dicer lox/lox纯合小鼠与CD4Cre阳性Dicer lox/lox纯合小鼠中分选得到CD4 T细胞;使用RNAbee试剂提取总RNA并完成扩增,随后将扩增产物与小鼠基因芯片进行杂交。每组设置3次生物学重复,分别对应3例Dicer野生型对照组与3例Dicer敲除组。
