We performed a forward genetic screening and identifid the transcription factor, salm. To identify potential downstream targets, we sorted the developing muscle cells from stage 11 embryo using GFP tagged by TEY-gal4 in wild type vs salm[340] mutants. This approach successfully identified the potential SALM-target genes. FACS-RNAseq of 2 Drosophila lines, with two technical replicates per line.

我们开展了正向遗传筛选,鉴定出转录因子salm。为鉴定潜在的下游靶标基因,我们利用TEY-gal4标记的绿色荧光蛋白(GFP),从野生型与salm[340]突变体的11期胚胎中分选发育中的肌细胞。该方法成功鉴定出潜在的SALM靶标基因。我们对2个果蝇品系开展了荧光激活细胞分选-转录组测序(FACS-RNAseq),每个品系设置2次技术重复。
