Foxd binding regions were examined by chromatin immunoprecipitation followed by deep-sequencing using 32-cell embryos of Ciona intestinalis (type A). In this experiment, a Foxd-Gfp fusion gene was expressed under the control of the upstream regulatory region of Foxd, and an anti-GFP antibody was used for immunoprecipitation.
本研究以A型玻璃海鞘(Ciona intestinalis)的32细胞期胚胎为实验材料,通过先进行染色质免疫沉淀(chromatin immunoprecipitation)再辅以深度测序(deep-sequencing)的实验策略,对Foxd结合区域进行了检测。本次实验中,研究人员将Foxd-Gfp融合基因(Foxd-Gfp fusion gene)置于Foxd基因的上游调控区域(upstream regulatory region)的调控下实现表达,并使用抗GFP抗体(anti-GFP antibody)完成免疫沉淀操作。
