A compressive mechanical force-induced-TMJ OA rat model was used in the study. Lenti-virus targeting RIP1 was constructed in vitro and injected into the TMJ. Hematoxylin and eosin staining, TUNEL staining and micro-computed tomography were used to assess histological and morphological changes in the mandibular condyle. Immunohistochemistry, Western Blot and real-time PCR were performed to determine the changes of inflammatory, apoptotic and necroptotic markers.

本研究采用压缩机械力诱导的颞下颌关节骨关节炎(TMJ OA)大鼠模型。体外构建靶向受体相互作用蛋白1(RIP1)的慢病毒(Lenti-virus),并将其注射至颞下颌关节内。采用苏木精-伊红染色(Hematoxylin and eosin staining)、TUNEL染色(TUNEL staining)及微型计算机断层扫描(micro-computed tomography)评估下颌髁突的组织学与形态学变化。通过免疫组织化学(Immunohistochemistry)、蛋白质印迹法(Western Blot)及实时荧光定量聚合酶链式反应(real-time PCR)检测炎症、凋亡及坏死性凋亡标志物的表达变化。
