This dataset presents an optimized AFLP experimental protocol for extremely large genomes exceeding 70 Gb. It uses restriction endonucleases AscI and SbfI that recognize 8-bp recognition sequences, employs primers with 3 selective bases to reduce the number of amplified fragments, and analyzes the effects of fragment quantity and genome size on homologous fragments (size homoplasy), as well as the impacts of different reaction conditions on experimental repeatability and band intensity in extremely large genome species such as mistletoe.
