Syntrophus aciditrophicus is a syntrophic bacterium that degrades key intermediates during anaerobic decomposition, such as, benzoate, cyclohexane-1-carboxylate, and fatty acids to acetate when grown with hydrogen/formate-consuming microorganism. In this study the transcriptomes of S. aciditrophicus grown in pure culture on crotonate, and coculture with Methanospirillum hungatei on crotonate, cyclohexane-1-carboxylate, and benzoate were sequenced using Illumina sequencing to determine whether or not a central metabolic pathway was reponsible for degradation of its substrates and formation of alicyclic and aromatic rings. In addition, transcriptomic studies were performed to deduce the enzymes responsible for ATP formation via substrate-level phosphorylation.

嗜食互营单胞菌(Syntrophus aciditrophicus)是一类互营细菌,在与消耗氢/甲酸的微生物共培养的条件下,可将厌氧分解过程中的关键中间产物——苯甲酸、环己烷-1-羧酸及脂肪酸——降解为乙酸。本研究采用Illumina测序技术,对以巴豆酸盐为唯一碳源的纯培养S. aciditrophicus,以及与洪氏甲烷螺菌(Methanospirillum hungatei)共培养、分别以巴豆酸盐、环己烷-1-羧酸和苯甲酸为碳源的S. aciditrophicus的转录组进行测序,以明确是否存在核心代谢通路负责底物降解以及脂环族环与芳香族环的形成。此外,本研究还开展了转录组学研究,以推导通过底物水平磷酸化途径生成ATP的相关酶类。
