Three technical lignins (hydrolysis lignin, HL; kraft lignin, KL; lignosulfonate, LS) were considered for the study. Lignins dissolved in M9-minimal media, along with a glucose (G) control, were batch cultivated with the gram-positive soil bacterium Bacillus licheniformis for 26 hours. Samples were taken in the mid-exponential phase of bacterial growth for transcriptome analysis. The primary goal of this study is to elucidate the genes involved in the utilisation of lignin-derived aromatics. Illumina paired-end sequencing has been performed to address this.

本研究选用三种工业木质素(technical lignin):水解木质素(hydrolysis lignin, HL)、硫酸盐木质素(kraft lignin, KL)及木质素磺酸盐(lignosulfonate, LS)。将上述木质素溶解于M9基础培养基(M9-minimal media)中,并设置葡萄糖(glucose, G)对照组,与革兰氏阳性土壤细菌地衣芽孢杆菌(Bacillus licheniformis)进行26小时的分批培养。在细菌生长的指数中期阶段采集样本以开展转录组分析。本研究的核心目标是阐明参与木质素衍生芳烃利用的相关基因,并通过Illumina双端测序(Illumina paired-end sequencing)完成相关测序工作以实现该研究目标。
