Addressing the still unresolved question of the existence of CIMP in breast cancer DNA methylation alterations among histological breast cancer subtypes were examined by applying the HM450K BeadChip of Illumina Inc., covering about 480,000 single CpG sites. In addition to the occurrence of the hypermethylator phenotype in breast cancer, its relation to genetic instability and histological subtypes was investigated. Bisulfite converted DNA from 32 fresh-frozen primary breast tissue samples and 4 breast cancer cell lines were hybridized onto the HumanMethylation 450K BeadChip from Illumina Inc. Additionally, 28 of these primary samples were analyzed by array-based comparative hybdridization (aCGH).

针对乳腺癌组织学亚型中,乳腺癌DNA甲基化改变相关的CpG岛甲基化表型(CIMP)是否存在这一尚未阐明的科学问题,本研究采用Illumina公司的HM450K芯片(HM450K BeadChip,覆盖约48万个单CpG位点)对相关亚型进行了检测。除探究乳腺癌中高甲基化表型的发生情况外,本研究同时对该表型与基因组不稳定性及组织学亚型之间的关联展开了研究。本研究将32例新鲜冰冻原发性乳腺组织样本与4株乳腺癌细胞系的亚硫酸氢盐转化DNA,与Illumina公司的HumanMethylation 450K芯片进行杂交。此外,本研究通过基于芯片的比较基因组杂交(array-based comparative hybridization, aCGH)技术,对其中28例原发性乳腺组织样本进行了分析。
