The goal of this project is to investigate whether we can use host-directed therapy (ruxolitinib) in conjunction with the anti-malarial drug artemether-lumefantrine to enhance immune responses in malaria-naïve individuals participating in a controlled human malaria infection model with P. falciparum-induced blood-stage malaria. With support from the Gates Foundation’s Collaboratory, we have generated Phospho CyTOF/mass cytometry data on WB PROT1 samples collected from study participants. The samples were stained in batches of plates. All the Stims for a given timepoint for a given study donor were stained together on the same plate as a Barcoded pool (CellID 20plex Standard Biotools). A healthy donor Unstim and IFNa+LPS+IL7 triple stim QC control was stained at the same time as each plate, but was barcoded and pooled as a separate Pool. The samples were normalized in the Standard Biotools EQ4 Passport method using Helios software, followed by debarcoding using the Helios software. The Debarcoded files are Ungated (still contain Beads, doublets, debris, etc). No live-dead was included in the staining, as the samples were initially fresh complete WB.
