Single-cell 5’ RNA-seq profiles of E16.5 forebrains in WT and Zfp661 knockout mice

In this assay, we aimed to investigate the impact of ZFP661 on the expression diversity of clustered protocadherin genes (Pcdhs). To accomplish this, we conducted single-cell 5’ RNA-seq analysis on developing mouse forebrains at embryonic day 16.5 (E16.5). Our findings reveal that ZFP661 enhances the expression diversity of Pcdhs by balancing the usage of Pcdh isoforms. We compared the usage of Pcdh isoforms and the expression diversity of Pcdhs between wild-type (WT) and Zfp661 knockout (KO) developing forebrains at E16.5. Two pairs of littermates, consisting of WT and Zfp661 KO mice, were utilized for the experiment.
本实验旨在探究锌指蛋白661(ZFP661)对成簇原钙粘蛋白基因(Pcdhs)表达多样性的影响。为达成该研究目标,我们对胚胎发育第16.5天(E16.5)的小鼠发育前脑开展了单细胞5’端RNA测序分析。研究结果显示,ZFP661可通过平衡原钙粘蛋白同工型的使用,提升Pcdhs的表达多样性。我们对比了胚胎发育第16.5天(E16.5)的野生型(WT)与Zfp661基因敲除(KO)小鼠发育前脑中的原钙粘蛋白同工型使用情况及Pcdhs表达多样性。本实验共使用2对同窝野生型及Zfp661基因敲除小鼠作为实验材料。
