Particle size is an important parameter
for assessing the “health status” of microbial cells. For example, cells
deficient in the cell division machinery may be concatenated into long
filaments. This dataset describes some initial observations of number-average
particle size of Bacillus subtilis NRS-762 (ATCC 8473) and genetically
engineered and wildtype Chromobacterium violaceum ATCC 12472 cultivated
in different growth media over different cultivation periods. At appropriate
timepoint, samples were pipetted from the culture broth into 15 mL polypropylene
centrifuge tube. Dilution was applied when necessary to profile for any
dilution related effect on particle size distribution. Vigorous shaking by hand
was used to disperse any cell clusters prior to particle size analysis, which
was carried out with the pipetting of sample into a square cuvette for analysis
in Malvern Zetasizer Nano ZS instrument.
